Kanamycin Aesculin Azide Agar is a selective isolation and enumeration medium for enterococci (Lancefield group D streptococci) in food, and is not intended for use in the diagnosis of disease or other conditions in humans.
Sodium azide and kanamycin provide the selective inhibition required whilst aesculin and iron salts form an indicator system for the presumptive identification of enterococci. Incubation at 42°C will increase the medium’s selectivity.
|Yeast Extract||5.0 g/L|
|Sodium Chloride||5.0 g/L|
|Sodium Citrate||1.0 g/L|
|Ferric Ammonium Citrate||0.5 g/L|
|Sodium Azide||0.15 g/L|
|Kanamycin Sulphate||0.02 g/L|
Final pH: 7.0 ± 0.2 at 25°C
Formula may be adjusted and/or supplemented as required to meet performance specifications.
- Suspend 43 grams of the medium in one 1 liter of purified water.
- Heat with frequent agitation and boil for one minute to completely dissolve the medium.
- Autoclave at 121°C for 15 minutes.
- Cool to 45-50°C.
